The Contractor will conduct basic and applied research on current and emerging threats at BSL2 and BSL3 levels, in support of the biodefense and infectious disease program at USAMRIID. The Contractor will provide expertise in a range of immunological assessments supporting prophylactic and therapeutic product development against botulinum toxin. The contractor shall provide laboratory support with laboratory experiments in BSL-2/3 CDC registered laboratories and thus must hold and maintain BPRP status. Responsibilities include:
- The Contractor will perform assessment of recombinant protein vaccine antigens in small animal biological assays. This will include vaccine formulation, inoculation of test subjects, administration of challenge toxins, and assessment and quantification of vaccine potency and efficacy. This includes approximately 15-20 animal experiments per year.The Contractor will perform assessment and characterization of immunotherapeutic molecules in both in vitro and in vivo models. This may include determination of antibody titers by ELISA, toxin neutralization assays, passive transfer studies, and assessment of individual immunoglobin preparations. The contractor will produce and characterize challenge toxins from strains expressing toxin variants not commercially available for use in vaccine challenge studies. This may include culturing select agent strains of pathogens, partially clarifying toxin extracts, determining toxin potencies and characterizing toxins by SDS-PAGE, Western blot, and UV spectroscopy.The contractor will characterize new Clostridium botulinum strains as to morphological, biophysical, and genetic characterizations. They will produce cell, spore, crude toxin supernatant, and extracted DNA stocks from C. botulinum cultures. Organize and maintain stocks for efficient inventory, archiving, and retrieval. Collect and organize data on C. botulinum strains.The Contractor will prepare animal protocols, standard operating procedures, and other documentation necessary to conduct assigned experiments. The contractor will be responsible for assessing and maintaining laboratory supplies in assigned laboratories. Will assess and ensure that supplies are ordered or will be ordered using the current GFEBS ordering system. Responsible for maintaining assigned laboratory space in compliance with all applicable regulations.The contractor will assist the Toxin Vaccine Research Scientific Support personnel The contractor will assist in the preparation of statements of work, annual reports and manuscripts related to their research and assigned tasks. The contractor should present relevant research data atone scientific meeting per year.Prepares monthly Toxin Vaccine Research Technical Reports and reports data results at laboratory meetings.
